Melzers reagent

Dried herbarium specimens are deposited at Oregon State University (OSC) with duplicates deposited at the University of Florida (FLAS). Specimens were hand-sectioned and then mounted in water and Melzer's reagent. We examined, measured, and photographed spores from each specimen and checked for an amyloid response in Melzer's reagent.

A microstructure reacts hemiamyloid when all the following apply: (1) direct application of Lugol’s solution (without KOH-pretreatment) provokes a red or red-brown reaction, (2) direct application of Melzer’s reagent yields no reaction at all (due to the high amount of chloral hydrate in that reagent), and (3) a blue reaction is obtained when th...Lugol's solution/IKI versus Melzer's reagent: Hemiamyloidity, a universal feature of the ascus wall. For microscopic studies, sections of dried basidiocarps were rehydrated in 3% KOH, subsequently stained in Congo red solution, Melzer's reagent and Cotton blue, and then observed under an Olympus BX 53 (Tokyo, Japan) light microscope (Nikon, Tokyo, Japan) at either 40, 100, 400, 600, and 1000 magnifications.Intact AM fungal spores were transferred using a wet needle to polyvinyl alcohol-lacto glycerol with or without Melzers reagent on a glass slide for identification. Spores were identified based on spore morphology and subcellular characters and compared with original descriptions (Schenck and Perez 1990 ).

A small part of each sporocarp was then scratched and mounted in polyvinyl alcohol lactoglycerol (PVLG; Omar et al. 1979) or a mixture of PVLG and Melzer's reagent (1:1, v/v). The resulting spore slides were examined under an Olympus BX51 microscope equipped with differential interference contrast.A 5-10 spores of each spore type were mounted in polyvinyl-lacto-glycerol (PVLG) and PVLG-Melzer reagent (1:1, v:v) on a glass slide; the spores in Melzer reagent were crushed in order to observe staining of different layers of spore wall. Spores were examined using a compound microscope (Olympus AX70) with Nomarski optics. ...
